I would like to know how effective the ANC in Odyssey is. What happens if the OEM stereo system is replaced with a custom audio system? Or, is there a way to keep the ANC functional if we keep the tuner?

If you get an after market stereo, ANC will be disabled, period. ANC minimizes low frequency "rumbles" from the engine and the road. If you turn it off the difference is clearly noticeable but its not huge. Personally I dont see how you can swap an odysseys OEM stereo without crapping the whle center stack since its a non standard unit. Furtheremore, the stereo is an integrated unit that controls the nav, phone etc etc, unless you have an LX with its simple radio I would not endeavor in aftermarket unit, of course you could always swap the speakers (as I did on our 99 LX) but that is mostly it. You can spend a lot of K's on a custom system, to make it the ultimate boom boom van but what 's the point? The touring vans have decent audio already.

Has anyone doing measured road test ever measured the noise levels with ANC on and off?

Don't know but in terms of dB its probably small 1-2 dB at most. Its mostly a "quality of sound" modifier. One way to check is to cover the cabin microphones; one in the front and one in the rear. I'll give it a try and see what happens. A cotton ball and some tape should do the trick. When my RL was new, subzero temps would give the mike issues the stereo would have a low level wheeee sound til things warmed up or i tapped it a couple of times, it went away on its own.

fyi, 3rd gen owners that changed out their speakers/subs (i forgot which one) noticed a droning from the speakers when ANC was activated.

If you swap your stock speakers for after markets. Have to toy with the wiring to make sure your new fronts are in phase with your new rears (and tweaters), connecting + to - and + to + and seeing which sounds better.

NO need to "play" with them. In my experience, the speaker binding posts have the plus or minus signs marked so all you have to do is remember which cable went where. The pain involved in speaker swapping is finding speakers that will fit well within the original housing so as to minimize cutting and drilling screws into where they were not supposed to. Its also a pain to remove panels that werent really meant to be removed and putting them back on there is always some clip or attachment that gets damaged in the process. Replacing the speakers on my s2K was a nightmare!

It's not the wiring, it's the speakers, different companies do it differently, so you may need to reverse the polarity on one set so the cones are "out" at the same time. I don't know the tech of it, but something like the current going through the coil in the opposite direction.
But you cannot rewire the speaker, so you need to rewire the wires.
